EpiTox – completing our ValidaTe platform for TCR-mimic antibody development

12.12.2025
Following our EpiPredict publication
(click to read the publication), we’re pleased to announce the pre-print release of EpiTox – completing our Validate platform for TCR-mimic antibody development.

As outlined in our EpiPredict paper, EpiTox is a machine-learning–powered platform for peptide-centric pHLA off-target risk assessment, addressing a critical safety challenge in TCR-mimic (TCRm) antibody development.

Using MAGE-A3 as a case study, we demonstrate how EpiTox identifies sequence-related, high-risk off-target peptides. Importantly, applying EpiTox to an anti–MAGE-A3 TCRm antibody revealed previously unrecognized off-targets, underscoring its value for early, mechanism-driven safety de-risking and robust candidate selection in pre-clinical development.

With the publication of EpiTox, all three components of our ValidaTe platform are now publicly validated and available:

  1. SCORE: proprietary high-throughput pHLA screening

  2. EpiPredict: TCRm-centric, proteome-wide off-target identification

  3. EpiTox: peptide-centric pHLA off-target risk assessment (Link to paper)

Looking ahead: Updates on our MAGE-A4 lead molecules are planned for Q1 2026.
